چکیده

Epigenetic mechanisms are known to be involved in tissue-specific differentiation. DNA methylation patterns have been shown to be largely conserved across tissues but with variation for specific genes. However, it is unclear whether the variability observed in the methylation profile of a metabolically active tissue is reflected in other sources such as hematopoietic tissue. This study aimed to test blood genome-wide CpG site methylation levels as a surrogate model for visceral adipose tissue (VAT) methylation and to verify whether it appropriately reflects differences in methylation levels found in VAT between men discordant for the metabolic syndrome (MetS). Tissue specimens (VAT and blood samples) were obtained from 16 severely obese individuals discordant for the MetS. CpG sites methylation levels were measured with the Infinium HumanMethylation450 BeadChip and correlations of methylation levels between VAT and blood were computed. Differences in methylation levels between individuals with and without MetS were tested in both tissues. Pathway analysis was conducted for differentially methylated CpG sites common to both tissues. High cross-tissue correlations were observed for VAT and blood (0.952±0.014) while some CpG sites had significantly different methylation levels in VAT versus blood. Differential methylation analysis between individuals with and without MetS demonstrated a higher number of differentially methylated CpG sites in VAT than in blood (11,778 vs. 881, respectively) with nearly 4% of differentially methylated sites found in VAT being also represented in blood. Common differentially methylated sites were involved in inflammatory-, lipidand diabetes-related pathways. These results suggest that blood methylation levels of specific CpG sites may adequately reflect VAT methylation levels for some of the MetS-related genes, specifically for inflammatory, lipid and glucose metabolism genes.

The Association of Omentin Gene Expression in Visceral and Subcutaneous Adipose Tissues with Plasma Fatty Acids Profile and Dietary Fatty Acids

Introduction: Omentin, an adipokine, with anti-inflammatory effects reduces insulin resistance, and can hence, play an important role in prevention of cardiovascular disease and diabetes. The present study aimed to investigate the association of plasma and dietary fatty acids with gene expression of omentin in visceral and subcutaneous adipose tissues. Materials and Methods: Visceral and subcut...

Echocardiographic Epicardial Adipose Tissue as a New Indicator of Cardiovascular Risk

Abstract Background and Purpose: Epicardial fat is the true visceral fat located around the heart, particularly around sub-epicardial coronary arteries that may locally interact and modulate the coronary arteries and myocardium through paracrine or vasocrine secretion of anti-inflammatory and proatherogenic cytokines. Most previous studies have used echocardiography and reported controversia...

بررسی‏ قطر LDL-c و سطح sdLDL-c سرم در ارتباط با فنوتیپ هیپرتری‏گلیسریدمی با دور کمر بالا در جمعیتی از زنان

Background and Objectives: Obesity is a major risk factor of cardiovascular diseases. Visceral adipose tissue (VAT) plays an important role in complications obesity. Although the measurement of waist circumference is a useful index for evaluating obesity and its complications, always increasing of waist circumference is not associated with VAT accumulation. HTGW (Hypertriglyceridemic waist circ...

The Effect of Spinach Hydro-alcoholic Extract on Resistin mRNA Expression in Visceral Adipose Tissue of Rats Exposed to Chronic Restraint Stress

Introduction: Resistin is a peptide hormone, secreted by adipose tissue. Increased secretion of resistin results in insulin resistance and metabolic syndrome. The aim of this study was to determine the effect of hydro-alcoholic spinach (Spinacia oleraceae L.) extract on the expression of resistin mRNA in the visceral adipose tissue of rats exposed to chronic immobilization stress. Materials and...

Background & Aims: Physical activity by modulating angiogenesis in adipose tissue, effects on the entire body metabolic profile. Therefore, the aim of this study was to investigate the effect of 12 weeks of moderate intensity continuous training on VEGF, PEDF and PAI-1 of visceral and subcutaneous adipose tissue in obese rats. Materials & Methods: 15 male Wistar rats were divided into 2 groups...
